How to utilize Twitter?

How do you make the most out of Twitter? I always see hashtags and stories of people who contacted agents via tweet. I have a completed manuscript and am wanting to use Twitter to reach out. What now?

For getting agents' attention you can take part in twitter pitch events such as #pitmad and #pitchwars. On the selected day you tweet your book pitch with the right hashtags and if an agent likes it, that's an invitation to query them. Look here for more information: https://twitter.com/PitchWars

Agents want to be pitched on Twitter only at certain times. In addition to PitMad there are lots of smaller pitch days for specific genres, specific categories, and some for specific authors (LGBTA authors or POC authors, etc).

For the rest of it, follow a bunch of agents and authors and reply to their tweets the same way you'd reply to anyone's, and if you aren't too pushy about it, they may begin to recognize and like you, and then remember you when you query them.
